ABSTRACT

As the rapid development of minimally invasive techniques, anesthesia, and enhanced recovery after surgery (ERAS), anorectal day surgery receiving more and more attention by improving efficiency of medical care while reducing cost and hospitalized infection. However, day surgery also faces the challenge of completing the whole process from patient admission to discharge within 24 hours. Therefore, establishing a reasonable and detailed day surgery process is the cornerstone to guarantee safe medical practice and patients satisfaction. National Clinical Research Center for Geriatric Disorders (Xiangya), together with China Ambulatory Surgery Alliance formulates the clinical practice guideline for anorectal day surgery 2019 edition. Here we make some interpretations of the guidelines on the detailed process of anorectal day surgery, including indication, preoperative examination, preoperative risk evaluation, health education, assessment of day surgery anesthesia and before leaving postanesthesia care unit (PACU), postoperative management, assessment of discharge and follow-up, for the convenience of various medical centers.

ABSTRACT

Time activity pattern refers to the time and behavior of people at different locations. The knowledge of time-activity pattern is essential for air pollution exposure assessment when direct personal exposure monitoring can't not be conducted, because air pollutant concentrations may vary significantly by location and activity. This review is focused on the survey method for human time-activity pattern and its application in the exposure assessment of air pollutants. After comparing the diary-reported trips and Global Positioning System (GPS) recording method, we believed that diary-reported trips were more reliable in describing comprehensive and detail records of the behavior and environmental circumstances while GPS-recorded trend to maintain sound information of time, location and transportations. Meanwhile, according to different subjects, the air pollution exposure model could be divided for population and individuals. There were great difference between two types of model in many areas, including the required information about time, activity and microenvironment.

ABSTRACT

Objective To evaluate the method of flow cytometric analysis of T lymphocyte subsets in bodyfluid of the pa-tients. Methods In ovder to get T lymphocytes by 3-colour staining method utilizing SSC/CD3Percp antibody,47 bronchoalveolar lavage fluid and 50 were tested by flow cytometery. Results 1)The percentage of Th is (67.98±7.41) % ,Ts(27.36±6.41) % and Th/Ts(2.60±0.64)in BALF with lung cancer,and the percentage of Th is (37.61±4.8)% ,Ts(67.97±4.20)% ,and Th/Ts(0. 47±0.10)in BALF with lung cancer respectively, and the Th cells in BALF with lung cancer were lower than those in the benign pulmonary disorder(P<0.05), and the Th/Ts ratio was significantly lower while the Ts cells were significantly higher in BALF of lung cancer(P<0.01), comparied with the benign pulmonary disorder; 2)The percentage of Th is (35.76±7.71) % and Ts(64.31 ±7.37) % and Th/Ts(0. 57 ± 0. 18) in BALF with lung cancer, and the percentage of Th is(49.27 ± 12.88) %, Ts(44.54 ± 9. 56) %,and Th/Ts is (1.13 ± 0.29) in BALF with benign pulmonary disorder. Con-clusion The immunity function in the patients with lung cancer was significantly decreased. Flow cytometric approach is valuable in analyzing T lymphocyte subsets in BALF.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the mechanisms for the cytopathic effect (CPE) of human cytomegalovirus (HCMV) in ECV304 endothelial-like cells.</p><p><b>METHODS</b>PCR and indirect immunofluorescence were used to detect HCMV infection by examining immediate-early (IE) gene and protein expression of the virus in ECV304 cells. Phase-contrast and electron microscopies were performed to observe the morphological changes of the infected and uninfected cells, and DNA ladder analysis and flow cytometry were carried out to study HCMV-induced cell apoptosis.</p><p><b>RESULTS</b>In HCMV-infected ECV304 cells, cytopathic effects were first observed at approximately 72 h post-infection. The cells with CPE changes exhibited detachment from the monolayer, cell rounding and shrinkage. The expression of the IE gene was detected. Chromatin condensation and nuclear fragmentation along with dramatic changes of the mitochondria were observed by electron microscopy at 96 h post-infection. Cellular DNA fragmentation was observed in the infected cells, which had cells apoptotic rates of 4.1% and 45.7% at 96 h and 144 h post-infection, respectively.</p><p><b>CONCLUSION</b>HCMV can induce apoptosis of ECV304 endothelial-like cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the pathological changes and morphological alterations of ECV304 cells after the infection by herpes simplex virus type 2 (HSV-2) in vitro.</p><p><b>METHODS</b>Passaged ECV304 cells were infected with HSV-2, TCID50 and morphological changes were observed by optical microscopy and tissue staining.</p><p><b>RESULTS</b>One day after HSV-2 infection, swelling, rounding, and increase of thickened cytoplasmic granules occurred in the ECV304 cells, and on day 2, cell fusion was observed with weakened nuclear staining.</p><p><b>CONCLUSION</b>ECV304 cells mostly undergo necrosis after HSV-2 infection without obvious evidence of cell apoptosis.</p>

ABSTRACT

The effects of two different sample labeling methods on background signal intensities for high-density 60mer oligonucleotide microarray were investigated. Peripheral blood samples from five disease and five control subjects were collected. Total RNA targets from peripheral blood mononuclear cells were extracted and labeled with RD-PCR protocol, which were hybridized to Agilent Human 1B oligonucleotide microarrays in a two-color comparative format. The positive control targets were labeled with the directly incorporated fluorescently-labeled dNTP labeling. The SPSS program was performed to test normality of the dataset, variance homogeneity between the groups, coefficients of variation (CV) and analysis of variance. The results showed that the background signal intensities of Cy3 channel were higher than those of Cy5 channel.
